In Punchy Pooch, the plot revolves around Champ, a dog who is coaxed into participating in a boxing match against "The Australian Bounder," a kangaroo. The stakes are high, with a reward of $500 for anyone who can last four rounds with the kangaroo. However, the kangaroo and its manager employ unscrupulous tactics to ensure their victory.

Doc, Champ's companion, devises various strategies to help him prevail in the ring. These include tricking the kangaroo into believing that Champ is a mother with a child and later convincing it that Champ has the measles. In the final round, Doc puts Champ on a unicycle, adding a twist to the match as he tries to outmaneuver his opponent.
