Thanuppu tells the story of Pratheesh and Treesa, a young couple who move to a beautiful village surrounded by hills and rivers. They are assisted by a villager named Prakasan as they settle into their new rented room. Pratheesh finds work as a painter, integrating into the local community, yet their presence sparks curiosity among the villagers.

As the story unfolds, the villagers grow suspicious of Pratheesh and Treesa, particularly due to their repeated inquiries about the river's location.
